Aufgabenstellung
Material A: The Effect of the Pesticide Azamethiphos
Salmon lice ( Lepeophtheirus salmonis) are small parasitic crustaceans. They infect the Atlantic salmon, live on its body surface and feed on mucous, skin particles and the fish’s blood. Affected fish have open wounds in the skin and anaemia 1. This can be lethal for the fish.
In salmon farming, the infestation of the fish can cause great economic losses. As one measure, the pesticide Azamethiphos is used in aquafarming, where it is added to the breeding tanks in small amounts (0.2 µg per litre). In order to investigate the effect of the pesticide on the nervous system of salmon lice, homozygous wild type salmon lice were treated with Azamethiphos for 24 hours. The activity of the enzyme acetylcholine esterase (AChE) was then measured in the treated wild type salmon lice (Figure 1).

Material B: Azamethiphos Resistance and its Genetic Causes
After many years of regular use of Azamethiphos in aquafarming, a widespread resistance to this pesticide was observed in salmon lice. While investigating the causes for the resistance, the ace1a gene was discovered in the louse. The gene codes for acetylcholine esterase.
Material C: Effectiveness of Azamethiphos in Different Lines of Salmon Lice
In salmon farming waters of Norway, salmon lice lines 2 which differed in the genotypes of the ace1a gene were found. The effectiveness of Azamethiphos on salmon lice of the three lines was investigated: three salmon lice lines were treated with this pesticide for 24 hours and the effects observed (Figure 2). Then, the activity of the enzyme acetylcholine esterase was determined in line 1 under the influence of Azamethiphos (Figure 3).

Lösung
Nach den offiziellen Korrekturhinweisen
mRNA sequence of the wild type allele (s-allele):
Lösung: 5' GGU-UGC-UCC-GUA ... GGG-AAU-UAU-UUC 3'
Amino acid sequence of the s-allele:
Lösung: Gly-Cys-Ser-Val ... Gly-Asn-Tyr-Phe
mRNA sequence of the resistant allele (r-allele):
Lösung: 5' GGU-UGC-UCG-GUA ... GGG-AAU-UAU-UAC 3'
Amino acid sequence of the r-allele:
Lösung: Gly-Cys-Ser-Val ... Gly-Asn-Tyr-Tyr
Type of mutation in triplet 235:
Lösung: stumme Mutation / silent mutation
Type of mutation in triplet 362:
Lösung: Missense-Mutation / missense mutation
Consequences of the mutations on the function of acetylcholine esterase:
Lösung: The silent mutation at triplet 235 does not alter the spatial structure or function of the enzyme. The missense mutation at triplet 362 leads to the incorporation of tyrosine instead of phenylalanine. This can alter the tertiary structure, particularly near or at the active site, preventing Azamethiphos from binding effectively. Consequently, AChE is no longer inhibited by the pesticide.
Genotype of Line 1:
Lösung: RR / rr
Genotype of Line 2:
Lösung: ER / RE / Rs / Sr
Genotype of Line 3:
Lösung: EE / SS / ss
Reasoning for genotypes of lines 1, 2 and 3:
Lösung: Line 1 shows high survival (low mortality ~15%), indicating homozygosity for the resistance allele (RR). Line 3 shows almost complete mortality (~96%), corresponding to homozygous wild-type sensitivity (EE). Line 2 shows intermediate mortality (~35%), which corresponds to a heterozygous genotype (ER) with intermediate resistance.
Compare the results in Figure 3 with those in Figure 1 and develop a hypothesis to explain the observed results:
Lösung: Comparison: In resistant salmon lice (Line 1, Fig. 3), AChE activity is also reduced by Azamethiphos, but to a lesser degree than in wild type (Fig. 1). Moreover, the baseline activity without pesticide is higher in resistant lice (mean ~72 vs. 60 units), and the remaining activity under 2 µg/L Azamethiphos is substantially higher (mean ~17 units vs. <10 units in wild type), with a broader range of variation. Hypothesis: The residual AChE activity of approx. 17 relative units in resistant lice is sufficient to terminate synaptic transmission at cholinergic synapses, preventing fatal continuous excitation. Additionally, the mutation in ace1a may confer a slight gain of function or higher baseline expression, maintaining crucial synaptic clearance.
